產(chǎn)品介紹 This gene encodes four products of the tachykinin peptide hormone family, substance P and neurokinin A, as well as the related peptides, neuropeptide K and neuropeptide gamma. These hormones are thought to function as neurotransmitters which interact with nerve receptors and smooth muscle cells. They are known to induce behavioral responses and function as vasodilators and secretagogues. Multiple transcript variants encoding different isoforms have been found for this gene. P物質(zhì)(Substance P)是腦組織及消化道中發(fā)現(xiàn)的一種神經(jīng)遞質(zhì)肽。由130個氨基酸殘基組成,胃腸道中肌肉收縮的有力促進劑,亦為強血管擴張劑。
SP是一種腦腸肽,亦是最早發(fā)現(xiàn)的一種神經(jīng)肽,屬于速激肽家族成員,它廣泛分布于中樞及外周神經(jīng)系統(tǒng),作為一種神經(jīng)遞質(zhì)或調(diào)質(zhì)參與疼痛、免疫、心血管等多種生理功能的調(diào)節(jié)。SP與其它的遞質(zhì)或調(diào)質(zhì)類似,能作用于多種類型的受體,速激肽家族有NK1、NK2、NK3三種受體,都能與SP結(jié)合,其中NK1與SP的結(jié)合能力最強而被稱為SP受體。

Paraformaldehyde-fixed, paraffin embedded (rat adrenal gland); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with ( Substance P) Polyclonal Antibody, Unconjugated (bs-0065R) at 1:200 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.
U-2OS cell; 4% Paraformaldehyde-fixed; Triton X-100 at room temperature for 20 min; Blocking buffer (normal goat serum, C-0005) at 37°C for 20 min; Antibody incubation with (Substance P) polyclonal Antibody, Unconjugated (bs-0065R) 1:100, 90 minutes at 37°C; followed by a conjugated Goat Anti-Rabbit IgG antibody at 37°C for 90 minutes, DAPI (blue, C02-04002) was used to stain the cell nuclei.
Blank control: Mouse brain cells (blue). Primary Antibody:Rabbit Anti- Substance P antibody(bs-0065R), Dilution: 1μg in 100 μL 1X PBS containing 0.5% BSA; Isotype Control Antibody: Rabbit IgG(orange) ,used under the same conditions ); Secondary Antibody: Goat anti-rabbit IgG-PE(white blue), Dilution: 1:200 in 1 X PBS containing 0.5% BSA. Protocol The cells were fixed with 2% paraformaldehyde (10 min). Primary antibody (bs-0065R, 1μg /1x10^6 cells) were incubated for 30 min on the ice, followed by 1 X PBS containing 0.5% BSA + 1 0% goat serum (15 min) to block non-specific protein-protein interactions. Then the Goat Anti-rabbit IgG/PE antibody was added into the blocking buffer mentioned above to react with the primary antibody at 1/200 dilution for 30 min on ice. Acquisition of 20,000 events was performed.
